2. Operation steps
1. Preparation
Prepare the required samples and reagents according to experimental needs to ensure that the instrument works smoothly. Check whether the ultrasonic extraction instrument is in a dry state to avoid water vapor entering the instrument and causing damage.
2. Sample processing
Add sample to the vial taking care not to exceed the marked line. Add appropriate amounts of solvents and surfactants to promote separation and dissolution of fixed substances. Silica gel and filter paper can be added to fix the active parts and plankton to avoid clogging the ultrasonic nozzle.
3. Instrument debugging
Place the sample bottle on the ultrasonic extraction instrument and adjust the frequency and power. The frequency is generally 20khz, and the power depends on the size of the sample and bottle. Adjust the time and temperature, usually 20-60 minutes, and control the temperature between 20-60°C.
4. Extraction process
After starting the extraction process, observe sample flow and solvent consumption. After the extraction is complete, stop the ultrasonic extractor and remove the sample vial. The sample is filtered and centrifuged in a centrifuge tube to remove unwanted material. Finally, transfer the liquid to a reaction bottle or other container for the next step.
